The proposed route will run along the A205 South Circular Road, operating as a single-deck bus service due to a low bridge on Thurlow Park Road.
Buses are expected to run every 12 minutes from Monday to Saturday between 5am and 12.30am, and every 15 minutes on Sundays, evenings and early weekday mornings.
TfL says the SL15 will reduce journey times and create faster connections between local centres, railway stations, and key work and leisure destinations.
The route will link with 77 other bus routes and eight train stations, including Tube, London Overground, and National Rail services.
It also aims to cut waiting times and ease congestion on nearby routes such as P13, 160 and 185.
