The bridge, which carries southbound traffic across the River Thames between Kent and Essex, will be shut from 10pm to 5.30am for essential maintenance.
Southbound vehicles will be diverted through the East Tunnel in a contraflow system.
This is part of a wider programme of overnight closures affecting both directions at the Dartford Crossing from Thursday, July 11 through to the early hours of Monday, July 14.
The crossing, used by over 150,000 vehicles each day, is one of the UK’s busiest road routes and a vital freight and commuter link between the southeast and the rest of the country.
National Highways said diversions will be clearly signposted via their network and local roads.
